چکیده

The age distribution of esterases in the interstitial cell of the mouse testis was studied between birth and the end of the tenth week of postnatal life by using the substrates O-acetyl-s-bromoindoxyl, a-naphthyl acetate, naphthol AS acetate, naphthol AS-LC acetate, and naphthol AS-D acetate. With indoxyl acetate, esterase activity is prominent at birth and wanes over the first 2 weeks of postnatal life. Esterase activity with the substrates a-naphthyl acetate, naphthol AS acetate, and naphthol AS-LC acetate is first demonstrable 2, 3, and 7 weeks after birth respectively, and in each instance increases steadily with age. Differences in the volume of reactive interstitial tissue for each of these substrates are •demonstrable at all ages. Naphthol AS-D acetate is only utilized by the interstitial cells of the 10-week mouse. No esterase substrate is used by all age groups, and quantitative differences at a given age exist between the individual substrates. These results are thought to imply the existence of at least 5 substrate-specific esterases in the mouse interstitial cell, and their possible relation to steroid synthesis is noted.

Postnatal differentiation and endocrine control of esterase isoenzymes in the mouse epididymis.

At 2 weeks of age, 11 isoenzymes were expressed and similar banding patterns on vertical polyacrylamide gel electrophoresis (PAGE), stained by alpha- or beta-naphthyl acetate as a substrate, were obtained for tissues or fluids from the proximal and the distal parts of the mouse epididymis. After this period, the emergence of new bands or the disappearance of certain others led to a regional dif...

Biochemical Characterization of A Novel Thermophilic Esterase Isolated from Shewanella sp F88

The main objective of this study was to purify and characterize an esterase from Shewanella sp F88. The enzyme was purified 41-fold and an overall yield of 21 %, using a two-step procedure, including ammonium sulfate precipitation and Q-sepharore chromatography. Molecular weight of the enzyme was 62.3 kDa according to SDS-PAGE data. The enzyme showed an optimum activity at pH 6.5 and 58 ˚C. Evo...

Identification of Spata-19 New Variant with Expression beyond Meiotic Phase of Mouse Testis Development

Background: The study of specific genes expressed in the testis is important to understanding testis development and function. Spermatogenesis is an attractive model for the study of gene expression during germ cell differentiation. Spermatogenesis associated-19 (Spata-19) is a recently-identified important spermatogenesis-related gene specifically expressed in testis. Its protein product is in...

The Role of Highly Conserved Tryptophan in the Sixth Conserved Region at Substrate Specificity of α- amylase

Early in this study, an α-Amylase from Bacillus megaterium WHO (BMW) was isolated from hot springs of Ramsar (North of Iran), and its gene was cloned in E.coli. Based on its conserved sequence regions and substrate specificity, it was classified as intermediary group enzymes with the specificity of oligo-1,6-glucosidase and neopullulanase subfamilies. In the sixth conserved re...

The hemagglutinin-esterase of mouse hepatitis virus strain S is a sialate-4-O-acetylesterase.

By comparative analysis of the hemagglutinin-esterase (HE) protein of mouse hepatitis virus strain S (MHV-S) and the HE protein of influenza C virus, we found major differences in substrate specificities. In striking contrast to the influenza C virus enzyme, the MHV-S esterase was unable to release acetate from bovine submandibulary gland mucin. Furthermore, MHV-S could not remove influenza C v...
